C minimal medium

This medium is used to grow B. subtilis in the presence of glucose or other variable carbon sources. The medium does contain ammonium and is therefore not suited to change the nitrogen source.

The basal solutions

  • 5 X C salts
    • 20 g KH2PO4
    • 80 g K2HPO4 X 3 H2O
    • 16.5 g (NH4)2SO4
    • Fill up to 1000 ml with deionised H2O
  • III’ salts
    • 0.232 g MnSO4 X 4 H2O
    • 12.3 g MgSO4 X 7 H2O
    • 12.5 ml of a ZnCl2 solution (100 mM)
    • Fill up to 1000 ml with deionised H2O
  • tryptophan (5 mg/ml)
  • Iron Ferric Ammonium Citrate (CAF; 2.2 mg/ml)
  • Potassium Glutamate (40 %)
  • Sodium Succinate (30 %)

Preparation of the medium

  • 1 X C minimal medium
    • 20 ml 5 X C salts
    • 1 ml Tryptophan (5 mg/ml)
    • 1 ml Iron Ferric Ammonium Citrate (CAF; 2.2 mg/ml)
    • 1 ml III’ salts
    • Fill up to 100 ml with deionised H2O
  • 1 X CSE minimal medium
    • 20 ml 5 X C salts
    • 1 ml Tryptophan (5 mg/ml)
    • 1 ml Iron Ferric Ammonium Citrate (CAF; 2.2 mg/ml)
    • 1 ml III’ salts
    • 2 ml Potassium Glutamate (40 %)
    • 2 ml Sodium Succinate (30 %)
    • Fill up to 100 ml with deionised H2O
  • 1 X CSE Glc minimal medium
    • is CSE minimal medium with 0.5% Glucose
  • 1 X CS minimal medium
    • is CSE minimal medium without Glutamate
  • 1 X CS Glc minimal medium
    • is CS minimal medium with 0.5% Glucose
  • 1 X C Glc minimal medium
    • is C minimal medium with 0.5% Glucose
